There are actually two ways to pierce his armor. The first way is to manually aim your pistol to consistently land headshots on El Tiburon. If you are successful enough, the armor will eventually break down, allowing your melee attacks to deal damage normally. From this point on, combat becomes much simpler.
Keep in mind that El Tiburon will continue to spawn additional enemies throughout the fight. It can be overwhelming, but it's also a steady source of ammo and healing items. You can also grab it as a human shield and reposition it to absorb incoming bullets.
The headshot strategy works reliably, but can take a while if your aim isn't perfect. Fortunately, there is a much faster way.
When the fight begins, make sure you have full health and are standing within close range of El Tiburon. Throw the smoke grenade directly at your feet and use rapid fire immediately, or shoot manually while the smoke is active. Doing so will cause a small explosion that will instantly shatter your armor, skipping the lengthy headshot process altogether.
Once his armor is destroyed, you can engage him normally using your sword. Keep blocking and counterattacking until your health bar is depleted. After that, El Tiburon will fall and you can continue the story and move on to the next chapter.
