Nightkin were born from the Master and created to be the pinnacle of their race. Known for their intelligence, strength, and pride, these super mutants were considered humanity's greatest evolution. Except that that peak was short-lived, as the Master's death scattered the few created Nightkins, forced to flee from the enemies who sought to kill them.
Nightkin's strength comes not just from their massive muscles, but from the stealth skills they provide. The Master gave them the Stealth Boys, which allowed them to sneak across the West Coast, but this technology provided addictive habits that weakened the Nightkin, giving them schizophrenia, brain damage, and incurable paranoia.
Appalachian Super Mutant

West Tek released FEVs into the Appalachian region even before World War I, but despite military intervention, most survived the nuclear hell environment. By 2102, the number of super mutants had dwindled, but the Enclave's involvement in the development of FEVs in the region led to further gains. This older variant is not as powerful as other variants. This is especially true if you're creating a standard super mutant with no special variants of note.
Because of the date it was created, the Appalachian Super Mutant can be considered the weakest variant in the entire American region. It is unlikely that these super mutants were not culled by the Brotherhood of Steel, who have occupied the region in the decades following the departure of the vault dwellers of Vault 76 to rebuild Appalachia, and there is no real difference in power between these variants, making them a greater threat than the average invader.
Commonwealth Super Mutants

Enter the Commonwealth, where super mutants are seen as a persistent threat that has outlived their creators. The Commonwealth's Super Mutants were born in the Institute, a boogeyman organization deep underground, using FEV samples discovered during research on Gen 3 synthesizers. Abandoned and returned to the surface, these super mutants are hostile and somewhat intelligent.
The Commonwealth's super mutants maintain their strength thanks to their sheer numbers, unique weaponry, and allies, including the mutant hounds they train. While this group's strength lies in sheer numbers, it is nonetheless considerably weaker than the other variants, as it was developed from a FEV sample rather than a full buffet. Nonetheless, these super mutants are not weak, and there is still a level of intelligence that may not be fully understood. However, this shows in the tactical territory they are willing to take and the limited speech they can have.

This lack of thought makes them dangerous, but the stupidity they present means there is no tactical genius to help them get through the few territories they claim through overwhelming numbers.

The West Coast Super Mutants were born on FEV-II in California, and were once the sole creations of the fanatic Master, who mutated himself beyond recognition in order to create an army of super mutants to transform humanity. Under the Master's rule, the Super Mutants ravaged the West Coast, forcing more humans to join for the sole purpose of ridding the world of war. Because super mutants will be improved humans who will have no reason to fight with each other.
Thanks to this strand of FEV and the genius of its Master, these super mutants possessed an intelligence that other mutants lacked, giving them an edge on the battlefield in their ability to think without cunning tactics and blind aggression. As such, the West Coast Super Mutants are still considered stronger, even decades after their Master's death, even though Unity and the military are permanently split.

There is only one Frank Horrigan, and his custom Power Armor makes him a threat like no other. Although he is the only Enclave Super Mutant, he is still a variant that overpowers all the remaining Super Mutants.
